摘要: 整合素作为异源二聚体跨膜糖蛋白复合物,通过介导细胞间、细胞 - 胞外基质以及细胞 - 病原体间的多维度生物通讯,在细胞黏附、增殖、凋亡及恶性肿瘤侵袭转移等病理生理过程中发挥重要调控作用。近年研究表明,整合素受体家族的表达异常或功能失调与多种肾脏疾病的发生密切相关,包括但不限于肾小球肾炎、肾小管间质病变、肾血管疾病、糖尿病肾病及多囊肾等肾脏疾病。本文综合最新研究进展,系统梳理整合素受体的分子结构特征、亚型分类体系、构象激活调控机制及其在不同肾脏疾病中的特异性作用模式,着重探讨其在肾组织微环境调控中的信号转导,旨在为解析肾脏疾病的分子机制和研发创新治疗策略提供理论依据。

Abstract: Integrins, functioning as heterodimeric transmembrane glycoprotein complexes, exert pivotal regulatory control over pathophysiological processes including cellular adhesion, proliferation, apoptosis, and malignant tumor invasion/metastasis through orchestrating multidimensional biological interactions among cell-cell, cell-extracellular matrix (ECM), and cell-pathogen interfaces. Emerging evidence demonstrates that aberrant expression profiles and functional impairments within the integrin receptor superfamily are mechanistically implicated in the pathogenesis of diverse renal pathologies, encompassing glomerulonephritis, tubulointerstitial lesions, renovascular disorders, diabetic nephropathy, and polycystic kidney disease. This review synthesizes cutting-edge research advancements to methodically delineate the molecular architecture, subtype classification paradigms, conformational activation regulatory mechanisms of integrin receptors, and their disease-specific operational modalities across heterogeneous renal pathologies. Particular emphasis is placed on elucidating their critical involvement in renal microenvironmental modulation via sophisticated signal transduction networks, while identifying prospective therapeutic targets. These insights aim to establish a conceptual framework for deciphering molecular pathogenesis and advancing innovative therapeutic strategies for renal-associated disorders.
