摘要:

焦亡(pyroptosis)是一种特殊类型的程序性细胞死亡,其特征是细胞肿胀、破裂,细胞内容物释放,导致强烈的炎症反应。促炎性是其区别于凋亡的最重要特征。焦亡发生机制是Nod样受体(Nlrs)等感受器识别细菌、病毒和宿主内源性分子及有毒外来物,诱发炎症小体的组装,从而引起下游半胱天冬酶1(caspase1)的激活,进而一方面切割gasdermin D(GSDMD)产生成孔性的GSDMDN以诱发焦亡,另一方面促进炎症因子白细胞介素1β(IL1β)及IL18释放,加重炎症反应。焦亡在缺血再灌注肾损伤、缺氧复氧损伤、糖尿病肾病、狼疮性肾炎等肾脏疾病的发生发展中起重要作用。

Abstract:

Pyroptosis is a special type of programmed cell death.It is characterized by swelling and rupture of cells,releasing of cellular contents,and a strong inflammatory response.Among those,its proinflammatory properties are the most important feature distinguishing it from apoptosis.The mechanism of pyroptosis is that receptors such as Nodlike receptors recognize bacteria,viruses and host molecules as well as toxic products,inducing assembly of inflammatory complexes,activating of caspase1.The activation of caspase1 cuts the gasdermin D (GSDMD) to produce poreforming GSDMDN inducing pore formation,and stimulates production of IL1β and IL18,aggravates inflammatory response.Pyroptosis plays an important role in kidney diseases such as ischemiareperfusion kidney injury,hypoxia reoxygenation injury,diabetic nephropathy and lupus nephritis.
