摘要: 急性肾损伤(AKI)是由多种因素导致的急性肾脏功能下降,常见病因包括缺血再灌注损伤、重症感染、肾毒性药物、急性尿路梗阻等。肾小管上皮细胞(TEC)线粒体分布集中、代谢活跃,对伤害刺激敏感,是AKI主要的受累细胞。紧密连接是连接相邻TEC的主要结构,避免了肾小管内物质通过细胞间隙自由渗漏,对维持TEC极性和屏障完整性具有重要作用。本文就AKI中肾小管上皮紧密连接的研究进展作一综述,分析紧密连接作为诊疗靶点在AKI领域的应用前景。

Abstract: Acute kidney injury (AKI) refers to the rapid decline in renal function by various factors, such as ischemia—reperfusion injury, severe infection, nephrotoxic drugs, and acute ureteral obstruction. Tubular epithelial cells (TEC) are the main cells affected in AKI, with a concentrated distribution of mitochondria, active metabolism, and sensitivity to injury stimuli. Tight junction, a major structure connecting adjacent tubular epithelial cells that prevents free leakage of substances in renal tubules across intercellular space, is critical for the polarity of tubular epithelial cells and the integrity of renal tubular epithelial barrier. In this review, we focused on the research progress on tight junction of renal tubular epithelial in AKI, as well as its potential role as a therapeutic target for AKI.
