核因子E2相关因子2调控糖尿病肾脏疾病中细胞稳态的研究进展

Advances of nuclear factor erythroid 2 related factor 2 regulation of cell homeostasis in diabetic kidney disease

摘要: 糖尿病肾脏疾病(DKD)是糖尿病最严重的并发症之一。其发病机制尚不明确,目前认为DKD受氧化应激和炎症等多种病理生理因素影响。核因子E2相关因子2(Nrf2)是一种内源性转录因子,其转录激活可促进下游抗氧化基因及细胞保护性基因表达,参与调节多种细胞功能,维持细胞稳态,在DKD进展中发挥重要细胞保护作用。近期研究发现,Nrf2调节细胞自噬,拮抗细胞凋亡和铁死亡,是氧化应激相关疾病,炎症和自身免疫性疾病的潜在治疗靶点。本文将综述Nrf2对DKD病理过程的调控作用新进展,探讨Nrf2治疗DKD的新机制。

Abstract: Diabetic kidney disease (DKD) is one of the most serious complications of diabetes mellitus. The pathogenesis of DKD remains unclear. Oxidative stress and inflammation are considered the main pathophysiological factors of DKD. As an endogenous transcription factor, nuclear factor erythroid 2-related factor 2 (Nrf2) activates the transcription of antioxidant genes and cytoprotective genes, involving in regulating various cellular functions and maintaining cellular homeostasis. Thus, Nrf2 plays an important cytoprotective role in the progression of DKD. Recent studies have shown that Nrf2 could regulate autophagy, and inhibit apoptosis and ferroptosis, which is a potential therapeutic target for oxidative stress-related diseases, inflammatory and autoimmune diseases. This article aims to review the recent research progress of the cryoprotection of Nrf2 pathway in DKD, and understand the potential mechanism of Nrf2 effect in DKD.
