摘要:

摘 要 脓毒症性急性肾损伤(AKI)发病机制复杂且缺乏有效的药物治疗。Toll样受体4(TLR4)是固有免疫系统中的病原模式识别受体之一,通过识别并结合革兰阴性杆菌细胞壁成分脂多糖(LPS),激活信号转导通路。TLR4信号通路激活后释放多种炎症介质如TNF-α、IL-1、IL-6介导脓毒症性AKI发生。因此,TLR4信号通路可成为脓毒症性AKI治疗的新靶点,已有TLR4信号通路阻断剂问世,有望提高脓毒症患者生存率。

Abstract:

ABSTRACT     One of the most frequent and serious complications in septic patients is acute kidney injury (AKI). AKI greatly worsens the already poor prognosis of sepsis and increases cost of care. To date, the therapies have been mostly supportive, there are no drugs that are FDA-approved to treat or prevent AKI. Toll-like receptor 4 (TLR4), the signal-transducing molecule of the lipopolysaccharide (LPS) receptor complex, plays a fundamental role in the sensing of LPS from Gram-negative bacteria. TLR4 mediates the actions of LPS through production and release of inflammatory mediators such as tumor necrosis factor and interleukin 6. Recent data suggests that interaction between Gram-negative bacteria and both systemic and renal-bound TLR4 may contribute to septic AKI. Consequently, TLR4 is a target of choice for the development of novel therapeutic interventions.
