姜黄素对急性腹膜炎大鼠腹膜Toll样受体2和6表达的影响

Effects of curcumine on expressions of toll-like receptor 2 and 6 in rat peritoneum during acute peritonitis

摘要:

摘 要  目的:研究在表皮葡萄球菌诱发的大鼠急性腹膜炎的过程中,姜黄素对Toll样受体2和6(TLR2,TLR6)的表达影响。 方法:采用腹腔注射表皮葡萄球菌制备大鼠急性腹膜炎模型,在制模前24h腹腔注射姜黄素[50 mg/(kg.d)],然后分别在造模后3h, 6h, 12h, 24h, 48h不同时间点处死实验动物并取材,RT-PCR法检测各组大鼠腹膜组织TLR2,TLR6的表达。分别通过Westen Blot、免疫组织化学技术检测各组核因子κB(NF-κB)的表达情况。Elisa法检测肿瘤坏死因子α(TNF-α)的表达情况。 结果:腹腔注射表皮葡萄球菌后,腹膜组织出现水肿和间皮细胞形态改变。RT-PCR结果显示模型组实验动物腹膜TLR2,TLR6的表达自感染后3h到24h出现时间依赖性升高。姜黄素处理组与模型组相比TLR2,TLR6的表达明显降低。与模型组相比,姜黄素处理组NF-κB,TNF-α的表达明显降低。此外,姜黄素处理组腹膜组织损伤较模型组为轻。 结论:姜黄素能够有效下调在表皮葡萄球菌诱发的急性腹膜炎早期免疫应答过程中TLR2,TLR6和TNF-α的过表达,其作用机制可能是通过NF-κB通路而发挥作用。

Abstract:

ABSTRACT  Objective: Curcumine has a long history of use in Chinese medicine as a treatment for inflammatory conditions. The purpose of this study was to investigate the preventive effects of curcumin against acute peritonitis induced by Staphylococcus epidermidis in rat and to elucidate possible mechanism of Curcumin action. Methodology: Curcumin [50 mg/(kg.d)] was intraperitoneally injected to rat for 24h, followed by injection of Staphylococcus epidermidis to induce acute peritonitis. After a survival time of 3, 6, 12, 24 and 48 h, TLR2, TLR6 RT-PCR was performed. The expression of NF-κB was analyzed by Western blot and immunohistochemistry, TNF-α level in rat peritoneal fluid was detected by ELISA.  Results: The histological examination with HE staining showed that the submesothelial zone became thickened with the edematous changes and obvious WBC infiltration in both parietal and viscera peritoneum. Curcumin treatment inhibited the elevation of expression of TLR2 and TLR6 in peritoneum of acute peritonitis rats. Both TLR2- and TLR6-RT-PCR were revealed identical patterns in peritoneum of acute peritonitis rats. The expressions both TLR2 and TLR6 were progressively enhanced in 3h, 6h, 12, 24 hours after infection. In Curcumin treatment group, there had a similar tendency for the expression of TLR2 and TLR6 in peritoneum of acute peritonitis rats, whereas the expressions were weakened in 6, 12, 48 after infection compared with model group. The expression of NF-κB at 24 hours after infection and a statistically was significant reduction in rat peritoneum as compared to that in model group. Staphylococcus epidermidis could significantly induce the expression of TNF-α in rat peritoneal fluid, which could be partially reversed by Curcumin. Conclusion: Our results suggest that Curcumin could attenuate peritoneum tissue injury by inhibiting the expression both TLR2 and TLR6 and the release of inflammatory cytokine TNF-α. These effects may involve downregulation of NF-κB.
