摘要: 器官芯片是一种微流体细胞培养装置,是医学、生物学、物理学等多个学科的交叉技术,旨在再现组织器官的结构功能,为体外实验提供一种新工具。近年来,多种器官芯片相继出现,在组织发育、药物筛选和疾病建模等领域展现独特的优势。在肾脏领域,研究重点集中在以肾脏芯片模拟肾单位的结构与功能,并在此基础上构建肾脏疾病的体外模型,研究其发病机制;或制备肾毒性模型,为新药开发提供新工具。本文通过回顾器官芯片的发展历史、组成及优缺点,分别介绍肾小管芯片、肾小球芯片、肾单位芯片及包含肾单位的多器官芯片的特点及应用,对器官芯片在肾脏疾病中的研究进展作一综述。

Abstract: Organonachip is a microfluidic cell culture device that integrates multidisciplinary technologies from medicine, biology, and physics. It aims to replicate the structure and function on organlevels and to provide a new tool for in vitro experiments. In recent years,several organonchips have been developed,showing unique advantages in the fields of tissue development,drug screening and modeling various diseases. With respect to nephrology, researchers focus on simulating the nephron with kidneyonachip to establish in vitro models of kidney diseases and explore the pathogenesis; or constructing nephrotoxic models for development of new drugs. This review will retrospect development, characteristics, advantages and disadvantages of organonachip, introduce features and applications of tubuleonachip, glomerulusonachip, nephrononachip, multiorgansonachip including kidney, and summarize research progresses of organonachip in kidney diseases.
