Abstract: Objective:To study the protective effect of resveratrol on contrastassociated acute kidney injury. Methodology:Twentyfour rabbits were divided into 4 groups (n=6): Control group (Control); Resveratrol group (Res); Contrastassociated acute kidney injury group (CAAKI); Resveratrol treatment group (CAAKI+Res). All rabbits underwent blood oxygen level dependent magnetic resonance imaging, R2* values of renal cortex (CO) and medulla (OM) were measured. Renal pathological changes, cleaved caspase3 and silent information regulator 2 homologous protein 1 (SIRT1), hypoxia inducible factor1α (HIF1α) and its downstream signaling pathways were estimated. Results:Compared with CAAKI group, R2* in OM (P<0001) and CO (P=0003) decreased significantly in CAAKI+Res group. Pathological injury scores showed that renal injury in CAAKI+Res group was less than that in CAAKI group (P<0001). Compared with CAAKI group, SIRT1 (P<0001), Bcl2 protein (P=0014) increased and HIF1α (P<0001), cleaved caspase3 (P=0018), Bax (P=0004) and CytC (P=0033) decreased in CAAKI+Res group. Conclusion:Resveratrol plays a protective role in CAAKI mainly through antihypoxia, antifibrosis and antitubular apoptosis. Blood oxygen level dependent magnetic resonance imaging technique can evaluate the protective effect of resveratrol on CAAKI.
