Effect of cell membrane mimetic polymer coating on biocompatibility of cellulose acetate membrane dialyzer

Abstract: Objective:To study the effect of cell membrane mimetic polymer PMNC coating on blood compatibility of dialyzer.
Methodology:PMNC polymer was coated on the surface of cellulose acetate (CA) membrane. The effects of membrane surface on fibrinogen(Fib) adsorption, adhesion and activation of platelets(PLT), complement and leukocytes were observed by in vitro protein adsorption and animal extracorporeal circulation (ECC).
Results:Compared with the uncoated CA membrane, the adsorption of serum bovine albumin and fibrinogen on the PMNC polymer coated CA membrane decreased by 81% and 82%, respectively. During the animal ECC, the PLT and WBC counts and the FIB concentrations in the uncoated dialyzer group decreased and the concentrations of βTG, IL8, C5a, TAT and F1+2 increased significantly. However, those parameters did not significant change during ECC through the PMNC coated dialyzers. After ECC 120 min, the coagulation of dialyzer was obvious in the uncoated group but no coagulation occured in the PMNC polymer group.
Conclusion:These results suggest that PMNC polymer coating could significantly improve biocompatibility of CA membrane dialyzer, which provides an important experimental basis for further applying PMNC polymer coating to the surface modification of medical devices.
