Double negative T cells in peripheral blood of patients with lupus nephritis: The relation to disease activity and therapeutic effect

Abstract: Objective:To investigate the proportion of double negative (DN)T cells in peripheral blood of patients with lupus nephritis (LN) and its correlation with disease activity and prognosis.
Methodology:A retrospective study was performed on patients with LN from 2010 to 2017.According to the median value of DNT cell proportion (460%), the patients were divided into low proportion group(n=138) and high proportion group(n=137).Clinicopathological characteristics and disease remission after 1 year followup of the two groups were compared.
Results:Of the 275 patients, 43 patients (156%) were male with a median age of 27 (21,37) years. The course of SLE and LN were shorter and the SLEDAI score (P=002) was higher in the high proportion group with higher level of lymphocyte proportion, uric acid, and antiC1q antibody, lower level of complement C3 and complement C4 (P<001). Patients in high proportion group had a lower chronic index(CI) score of renal lesions, globular sclerosis and crescent (P<005), and higher incidence of capillary thrombosis (P<005).The percentage of DNT cells decreased from baseline in remission group (P=0001), but not decrease in nonremission group (P=0954). There was no difference in remission rate between the two groups at 1 year of treatment.
Conclusion:The proportion of DNT cells in peripheral blood of LN patients correlated with disease activity of SLE, and decline of DNT cell ratio after treatment was associated with LN remission.
