Experimental establishment of cardiorenal syndrome type Ⅳ model

Abstract:

Objective:To investigate the viability of establishing stable model of cardiorenal syndrome type Ⅳ.
Methodology:(1) 7/8 nephrectomy were performed on Sprague Dawley rats for chronic kidney disease model,blood pressure,serum creatinine,serum urea nitrogen and creatinine clearance rate were monitred,Masson staining was used to observe the fibrosis of renal tissue.(2) Cardiac morphology at 1,3,and 5 weeks after nephrectomy was observed by echocardiogram.(3) The cardiac tissue was selected for wheat germ agglutinin staining,Masson staining and the expression measurement of cardiac hypertrophy indicators.
Results:5 weeks after 7/8 nephrectomy,rats in surgery group showed significant rise in blood pressure,serum creatinine and blood urea nitrogen,accompanied by decreased creatinine clearance rate and interstitial fibrosis in remnant kidney,serum phosphorus and fibroblast growth factor 23 were increased as well.Left ventricular myocardial wall thickening and mass index increased significantly,the expression of cardiac hypertrophy indicators (atrial natriuretic peptide and brain natriuretic peptide),myocardial interstitial fibrosis and myocyte cross section area were markedly increased.
Conclusion:
5 weeks after 7/8 nephrectomy,the stable model of cardiorenal syndrome type Ⅳ can be successfully established in Sprague Dawley rats.
